Rôle de l’innervation locale dans la formation des structures lymphoïdes tertiaires au sein des tumeurs pulmonaires non-à-petites cellules

Abstract : Tertiary lymphoïd sturctures (TLS) look like lymph nodes and share similar mechanisms of genesis. The involvement of the peripheral nervous system (PNS) in the genesis of lymph nodes has been suggested, however for TLS genesis it has not been documented. We hypothesized that the PNS is involved in the neogenesis of TLS. The objectives of my work have been to study the role of sympathetic nerve fibers in the formation of TLS using murine tumor and non-tumor models of pulmonary inflammation, as well as the characterization of fibers in patients' lung tumors.The destruction of sympathetic nerve fibers in both models (transient inflammation by lipopolysaccharide and tumor) is accompanied by a major reduction in the number of TLS in the denervated lungs compared to control mice. By 3D imaging, the vascular network, immune cells and sympathetic nerve fibers have also been observed in lung tumor biopsies.All of these data indicate that 1) sympathetic nerve fibers are present in patients' lung tumors; 2) the depletion of these fibers has a profound impact on the formation and size of TLS in the murine models studied.
